Running head: Raman imaging of secondary plant cell walls
نویسندگان
چکیده
Confocal Raman microscopy was used to illustrate changes of molecular composition in secondary plant cell wall tissues of poplar wood. Two–dimensional spectral maps were acquired and chemical images calculated by integrating the intensity of characteristic spectral bands. This enabled direct visualization of the spatial variation of the lignin content without any chemical treatment or staining of the cell wall. A small (0.5 μm) lignified border towards the lumen was observed in the gelatinous (G-) layer of poplar (Populus nigra x P. deltoids) tension wood. The variable orientation of the cellulose was also characterized, leading to visualization of the S1 layer with dimensions smaller than 0.5 μm. Scanning Raman microscopy was thus shown to be a powerful, non-destructive tool for imaging changes in molecular cell wall organization with high spatial resolution. www.plantphysiol.org on October 23, 2017 Published by Downloaded from Copyright © 2006 American Society of Plant Biologists. Raman Imaging of Plant Cell Walls in Sections of Cucumis sativus
Raman microspectra combine information on chemical composition of plant tissues with spatial information. The contributions from the building blocks of the cell walls in the Raman spectra of plant tissues can vary in the microscopic sub-structures of the tissue. Here, we discuss the analysis of 55 Raman maps of root, stem, and leaf tissues of Cucumis sativus, using different spectral contributi...
متن کاملIn situ label-free imaging of hemicellulose in plant cell walls using stimulated Raman scattering microscopy
BACKGROUND Plant hemicellulose (largely xylan) is an excellent feedstock for renewable energy production and second only to cellulose in abundance. Beyond a source of fermentable sugars, xylan constitutes a critical polymer in the plant cell wall, where its precise role in wall assembly, maturation, and deconstruction remains primarily hypothetical. Effective detection of xylan, particularly by...

متن کاملA Versatile Click-Compatible Monolignol Probe to Study Lignin Deposition in Plant Cell Walls
Lignin plays important structural and functional roles in plants by forming a hydrophobic matrix in secondary cell walls that enhances mechanical strength and resists microbial decay. While the importance of the lignin matrix is well documented and the biosynthetic pathways for monolignols are known, the process by which lignin precursors or monolignols are transported and polymerized to form t...
متن کاملIn situ micro-spectroscopic investigation of lignin in poplar cell walls pretreated by maleic acid
BACKGROUND In higher plant cells, lignin provides necessary physical support for plant growth and resistance to attack by microorganisms. For the same reason, lignin is considered to be a major impediment to the process of deconstructing biomass to simple sugars by hydrolytic enzymes. The in situ variation of lignin in plant cell walls is important for better understanding of the roles lignin p...
